MycoMonash Test

This test was developed by the Monash Tissue Culture Laboratory because regular and sensitive testing is essential for all cell and tissue culture laboratories.

The MycoMonash Test has been approved by Therapeutic Goods Administration (TGA licence No 15585).

The test is offered as an expert Mycoplasma testing service to other laboratories for research purposes as well as quality control of biotechnological manufacturing.

Mycoplasma Contamination

A major problem occurring in cultured cells. In some studies >25% of cultures examined were Mycoplasma positive

  • Mycoplasmas are extracellular parasites usually attached to the external surface of host cells.
  • Can be introduced through culture reagents, laboratory personnel and cross-contamination from other cultures.
  • Most common species: M. orale, M. hyorhinus, M. arginini, A. laidlawii, M. salivarium, M. hominis and M. fermentans.
  • Cell cultures are ideal for Mycoplasma parasite.
  • Microscopically undetectable.
  • Standard antibiotics (penicillin/streptomycin) do not prevent contamination.

Test Requirements and Characteristics.

PCR methodology is applied.

  • Only 10,000 cells required. Cells can be fresh, frozen or lyophilised.
  • Serum or culture medium.
  • DNA extract.
  • Sensitivity: A wide spectrum of Mycoplasma can be detected.
    Four organisms/sample can be detected.
  • Controls: Internal control is included as well as positive and negative controls.
  • Results can be available within 24 hours .
